Reversible epidermal growth factor receptor (EGFR) inhibitors depend on continued occupancy of the ATP-binding site. Covalent inhibitors can remain attached after unbound compound is removed. This makes exposure time and washout conditions important when comparing their effects: persistent inhibition of existing receptors does not necessarily mean permanent suppression of cellular signaling.

A Cyanoquinoline With a Reactive Side Chain
The EGFR inhibitor Pelitinib, also known as EKB-569, combines a 3-cyanoquinoline core with an anilino substituent at position 4 and a reactive butenamide chain at position 6. Its formula is C24H23ClFN5O2, with a molecular weight of approximately 467.9 g/mol.
The core engages the ATP pocket, positioning the unsaturated amide to react with a nearby cysteine. This Michael acceptor enables covalent attachment. Recovery of receptor activity then depends partly on protein turnover, rather than simply on inhibitor dissociation.
Potency and Selectivity
A reported biochemical IC50 for EGFR is 38.5 nM. Weaker activity was measured against Src at 282 nM and ErbB2 at 1.255 µM. These results indicate an EGFR preference within the tested panel, not exclusive activity against one kinase.
Cellular proliferation IC50 values were 61 nM in normal human keratinocytes (NHEK), 125 nM in A431 and 260 nM in MDA-468 cells, compared with 3.6 µM in MCF-7 cells. The response in normal keratinocytes shows that growth inhibition is not restricted to tumor cells. Proliferation measurements also differ from direct measurements of receptor phosphorylation.
Downstream Effects
In A431 and NHEK cells, reported signaling effects included:
- inhibition of EGF-induced EGFR phosphorylation at 20-80 nM;
- inhibition of STAT3 phosphorylation at 30-70 nM;
- reduced AKT and ERK1/2 activation at 75-500 nM.
The same study found no inhibition of the NF-κB pathway under its conditions. The different concentration ranges show why one downstream marker cannot stand in for every EGFR-dependent response. Studies in selected liver cancer cell models also reported growth inhibition around 2 µM, illustrating the importance of cellular context.
Results in Animal Models
In A431 xenografts, one oral 10 mg/kg dose reduced EGFR phosphorylation by 90% at one hour and by more than 50% at 24 hours. This persistence is consistent with irreversible inhibition, although the time course alone does not prove covalent binding.
At 20 mg/kg/day, the compound reduced tumorigenesis by 87% in APCMin/+ mice and limited virus-associated airway remodeling in a separate mouse model. These findings address distinct biological outcomes.
Pelitinib reached Phase II testing and serves as a research tool for EGFR signaling. Reported stock solubility is 20 mg/mL in DMF and 10 mg/mL in DMSO, with limited aqueous solubility. Final assay conditions require separate verification.
